The area that is now Canaseraga was first settled in 1808 by a group of settlers from Connecticut. The village was originally known as "Canaseraga Creek" and was later changed to "Canaseraga" in 1820. The name is derived from the Seneca Indian word for "long rapids".
The village was incorporated in 1848 and was the first village in Allegany County to be incorporated. The village was a major stop on the Genesee Valley Canal, which ran from Rochester to Olean. The canal was completed in 1851 and provided a major transportation route for goods and people.
The village was also home to a number of industries, including a sawmill, a grist mill, a tannery, and a woolen mill. The village also had a number of stores and shops.

There were 224 housing units at an average density of 363.2 per square mile (140.3/km2). The racial makeup of the village was 97.3% White, 0.6% African American, 0.2% Native American, 0.2% Asian, 0.4% from other races, and 1.2% from two or more races. Hispanic or Latino of any race were 0.6% of the population.
There were 206 households, out of which 33.0% had children under the age of 18 living with them, 51.5% were married couples living together, 11.2% had a female householder with no husband present, and 33.5% were non-families. 28.2% of all households were made up of individuals, and 14.1% had someone living alone who was 65 years of age or older. The average household size was 2.50 and the average family size was 3.06.
In the village, the population was spread out, with 27.2% under the age of 18, 8.3% from 18 to 24, 28.2% from 25 to 44, 21.7% from 45 to 64, and 14.6% who were 65 years of age or older. The median age was 36 years. For every 100 females, there were 91.2 males. For every 100 females age 18 and over, there were 87.2 males.
