Woodhull was founded in 1836 by a group of settlers from New York. The village was named after the first mayor, William Woodhull. The first post office was established in 1837.
In 1854, the Rock Island and Mercer County Railroad was built through Woodhull, connecting it to the larger cities of the region. This allowed for the growth of the village, and by the late 19th century, Woodhull had become a thriving agricultural community.
In the early 20th century, Woodhull was home to several businesses, including a flour mill, a creamery, a brickyard, and a furniture factory. The village also had a number of churches, schools, and a library.
