Pattonsburg was founded in 1837 by William Patton, a settler from Kentucky. The town was named after him and was incorporated in 1867. The town was a stop on the Hannibal and St. Joseph Railroad, which was the first railroad to cross the state of Missouri. The railroad was instrumental in the growth of the town, as it allowed for the transportation of goods and people.
